Which section of the policy is the setting which you have set? Is it a User Configuration or a Computer Configuration setting?
What objects are in the OU that the policy is applied to, a user or a computer?
Have you tried the GP Results section of the GPMC? This will list all policies and tell you the winning policy and if a policy fails it will give the reason why.
I am using User Configuration
Objects like removing the RUN and applying a wallpaper
Have you gone to Group Policy Management Console and set up a Group Policy Results test?

Miracle came in this morning and the policy that i applied last night worked. But when i change the policy again it dont get applied. I know it takes time to get implemented. But that is a joke!!!
What have you set under the following Domain GPO?
Computer Configuration/Administrative Templates/System/Logon/Always wait for the network at computer startup and logon

I have had a thought and yes that was my brain ticking you could hear.
In group policy have you set it so that it will wait for the domain or does it allow the machines to boot without talking to the server.
When applying group policy it says every 20 mins or so they should be updated on the client machine but at Carr Hill we average about 24 hours lols.
When working with Microsoft you have to wwwwwwwwaaaaaaaaiiiiiiiittttttt lols.
