You lose Single Instance Storage with Exchange 2010.
Depending on the amount of duplicate data in the databases this may or may not have a big effect.
Personally I allowed for A LOT more storage with 2010, but we've also moved to DAGS so have replicated databases, etc. Storage speed is less of an issue with 2010 too, so more, cheaper disk is an option.
1. Consider using the archiving feature of Exchange 2010 for the mailboxes or a 3rd party tool.
2. Consider implementing mailbox quotas
3. Consider Retension Policies/MRM
4. Yes, it will increaae but it depends on what's already in your databases.
