Is MS Course 6425A going to be the most relevant course for Group Policy?

Well, for a 2008 environment, Yes. There are slight differences between group policy in Windows 2003/R2 and Windows 2008/R2.

I learnt all from classes, on the job and books/CBT's. Group Policy & AD are very large subjects.
